PhD Mid-term Seminar Series: Infrared scattering type scanning near field optical microscopy (s-SNOM) in liquid **Infrared scattering type scanning near field optical microscopy (s-SNOM) in liquid** “Mid-infrared (IR) spectroscopy is a widely used label-free tool for secondary structure analysis and material identification in chemistry, biology and biochemistry. However, the diffraction limit of light prevents nanoscale resolved studies of proteins. Infrared scattering-type near-field optical microscopy (s-SNOM) is a scanning probe microscopy (SPM) technique which extends the power of mid-IR spectroscopy deep into the subwavelength regime, making it a highly sensitive tool for probing and mapping of protein secondary structure with nanoscale resolution in air. It is highly desirable to study biological samples in their native aqueous environments. However, the sensitivity of these techniques deteriorates dramatically in liquid environments. In this talk we will demonstrate a technique to enable nanoscale resolved chemical imaging and spectroscopy in liquid media.
